چکیده

The sea louse Caligus rogercresseyi is a marine ectoparasite that constitutes one of the major threats to salmon farming industry, where primary control strategy use delousing drugs through immersion treatments. emergence pharmacological resistance in this copepodid species has previously been described using transcriptome data. However, molecular mechanisms underlying chromosome rearrangements have not yet explored. This study aimed identify structural genomic variations and gene expression C. associated with pesticide sensitivity. In study, genome resequencing was conducted Oxford Nanopore Technology on lice strains contrasting sensitivity azamethiphos detect duplications. Transcriptome profiling putative duplications performed by Illumina sequencing. Copy Number Variants (CNVs) were identified comparative coverage, collinear/tandem over all chromosomal regions sequence homology. Duplications or CNVs functional genes primarily transposable elements related drug response, differential values calculated RNA-seq analyses same strains. Notably, differentially duplicated found coding cuticle proteins, suggesting mechanism may be cuticular structure formation proteins involved. Collectively, results revealed intensive pesticides populations increases frequency duplication, expanding involved response. first report an association between populations.

O-38: Concurrent Whole-Genome Haplotyping and Copy-Number Profiling of Single Cells

Background Methods for haplotyping and DNA copynumber typing of single cells are paramount for studying genomic heterogeneity and enabling genetic diagnosis. Before analyzing the DNA of a single cell by microarray or next-generation sequencing, a whole-genome amplification (WGA) process is required, but it substantially distorts the frequency and composition of the cell’s alleles. As a conseque...

Cathepsin Gene Family Reveals Transcriptome Patterns Related to the Infective Stages of the Salmon Louse Caligus rogercresseyi

Cathepsins are proteases involved in the ability of parasites to overcome and/or modulate host defenses so as to complete their own lifecycle. However, the mechanisms underlying this ability of cathepsins are still poorly understood. One excellent model for identifying and exploring the molecular functions of cathepsins is the marine ectoparasitic copepod Caligus rogercresseyi that currently af...

Disease Resistance in Atlantic Salmon (Salmo salar): Coinfection of the Intracellular Bacterial Pathogen Piscirickettsia salmonis and the Sea Louse Caligus rogercresseyi

BACKGROUND Naturally occurring coinfections of pathogens have been reported in salmonids, but their consequences on disease resistance are unclear. We hypothesized that 1) coinfection of Caligus rogercresseyi reduces the resistance of Atlantic salmon to Piscirickettsia salmonis; and 2) coinfection resistance is a heritable trait that does not correlate with resistance to a single infection. M...

The genomic architecture of segmental duplications and associated copy number variants in dogs.

Structural variation is an important and abundant source of genetic and phenotypic variation. Here we describe the first systematic and genome-wide analysis of segmental duplications and associated copy number variants (CNVs) in the modern domesticated dog, Canis familiaris, which exhibits considerable morphological, physiological, and behavioral variation. Through computational analyses of the...
